Tumörsuppressorn p53 som måltavla för ny cancerbehandling – Klas Wimans team

Vi bedriver forskning inom molekylär cellbiologi, tumörbiologi och nya cancerläkemedel. Vårt huvudintresse är tumörsuppressorn p53, en DNA-bindande transkriptionsfaktor som aktiveras som svar på DNA-skada och onkogen stress. Målet för vår forskning är att förstå hur p53 fungerar och skapa nya cancerläkemedel som återställer p53-funktionen i tumörer.

Tumörsuppressorgenen TP53

p53 reglerar cellulära processer som bl.a. cellcykelprogression, apoptos och metabolism genom att aktivera transkription av specifika målgener, t.ex. p21, Bax, Puma och Zmat3. Mutation i TP53-genen förekommer i omkring hälften av alla tumörer. De flesta TP53-mutationer är missense-mutationer som resulterar i förlust av p53s specifika DNA-bindning och förmåga att aktivera transkription. En mindre andel tumörer har nonsense-mutationer i TP53 som resulterar i ett trunkerat och icke-funktionellt p53-protein.

Reaktivering av muterat p53: en ny strategi för cancerbehandling  

Den höga frekvensen TP53-mutationer i tumörer gör att muterat p53 är en intressant mÃ¥ltavla för ny cancerbehandling. Vi har tidigare identifierat substansen PRIMA-1 och strukturanalogen APR-246 (PRIMA-1Met) som utlöser celldöd i tumörceller med TP53-mutation och hämmar tillväxt av tumörer in vivo i möss (Bykov et al., Nature Med. 2002; Bykov et al. Oncogene 2005). PRIMA-1 och APR-246 konverteras till den aktiva substansen MQ, en Michael acceptor som binder kovalent till cysteiner i p53s core-domän och ökar core-domänens termostabilitet (Lambert et al. Cancer Cell 2009;  Zhang et al. Cell Death Dis. 2018). Vi har ocksÃ¥ funnit att APR-246, via MQ, kan pÃ¥verka redoxbalansen i celler genom att hämma thioredoxin-reduktas och binda till glutathion (Peng et al. Cell Death Dis. 2013; Mohell et al. Cell Death Dis. 2015; Ceder et al. EMBO Mol. Med. 2021). APR-246 har testats i flera kliniska studier sponsrade av Aprea Therapeutics (Lehmann et al. J. Clin. Oncol. 2012; för en översikt, se Tuval et a. Nature Rev. Clin. Oncol. 2024). Vi utvecklar ocksÃ¥ ny behandling för tumörer med nonsense-mutation i TP53 genom farmakologisk induktion av translationell genomläsning. Vi har funnit att 5-FU-metaboliten 5-fluorouridine inducerar genomläsning och uttryck av aktivt full-längds-p53 i tumörceller med nonsense-mutationen R213X i TP53 vilket resulterar i celldöd via apoptos (Palomar-Siles et al. Cell Death Dis. 2022).

Zmat3 (Wig-1), en p53-målgen

Identifiering och karakterisering av nya p53-reglerade gener är viktigt för att förstå p53-signalvägen och p53-medierad tumörsuppression. Vi har tidigare identifierat den p53-reglerade genen Zmat3 (Wig-1) på kromosom 3q26.3-27 (Varmeh-Ziaie et al. Oncogene 1997; Hellborg et al. Oncogene 2001). Genen kodar för ett konserverat zinkfinger-protein med affinitet för dubbelsträngat RNA (Mendez-Vidal et al. Nucl. Acids Res. 2002). Våra resultat tyder på att Zmat3 styr p53-svaret mot cellcykel-arrest via reglering av specifika p53-målgener på mRNA-nivå (Bersani et al. Oncogene 2014).
